The underlying joke that led to the meme in the first place. That's usually how these things are created. A meme by itself has no meaning. Show somebody a meme they've never run across and they probably won't get the joke. Show it to somebody in the know and they'll laugh.

For example the Rick Roll wasn't the joke or the punchline for that matter. It was the end result of the joke. The joke was getting somebody to click on your link. The joke was hiding the truth from them by having it seemingly go elsewhere and then come around to Rick Astley. These people are acting like Kekistan is the punchline. It's not. They're the punchline because they don't get what it stands for.
