In Dark Souls 2, it's unquestionably the fuckers with the greathammers in the Dragon Shrine.

I haven't played through the entirety of the first game yet, but I've gotten to Blighttown and it is pure suffering.

The good thing about Blighttown is the blowgun enemies don't respawn after you kill them.

As well you can bypass the crypts and blighttown entirely by going through the Valley of Drakes through New Londo from the Firelink Shrine. It takes you just outside Quelaag's boss room. It's the way you can ring Quelaag's bell before the gargoyles.
 
Can bosses count? I want to say the Four Kings. I honestly despised fighting them at New Game + and beyond. It becomes more of a race than before in trying to kill one plus I really just find it to be a cheap boss. The Royal Rat Authority is another if simply because he has some wolf rats to back him up, which can cause toxic to players.

RRA was bullshit. Never finished my NG+ of DS1 because DS2 was right around the corner, but I can see how 4 kings could devolve into that. But yeah, RRA's minions were dumb. Royal Rat Vanguard was actually pretty interesting, though. I liked that bossfight a lot.

RRA is a Sif clone. It doesn't need flunkies to help it out (thankfully, it is an optional boss). I do feel it may as well be like 4 Kings in being the kind that gets help but thankfully, said help only respawns if you die rather than every thirty seconds (or how ever long it takes for a new king to spawn).
 
Skeleton Wheels forever the skeleton wheels. I've been killed by them so many time in my first playthrough it was ridiculous. I got stuck in the painted world because I couldn't see the lever in the basement.
The 4 kings suck if you can't do enough damage to kill one before the next spawns. Fighting 4 bosses at once in an environment that makes it hard to judge distances is a nightmare.

How do you think the skeleton wheels compare to the ice hedgehogs in the ivory king DLC though? They're basically skeleton wheels that 1 shot you and do damage to you if you get close to them and don't spin into walls forever.

Also, I think part of the reason I never had problems with the Kings is that I played faith w/ a divine claymore my first time through. That shit helps so much.

I was doing the Ivory King DLC on NG so I was bit over leveled so they weren't much of a problem and I generally tried to kill them with my bow first.
My first character was a physical knight character because I generally don't use magic in games. I also didn't maximize him so I was asking for trouble.
 
I never really had any trouble with The Four Kings, but I did always have a friend to help.

I voted for skeleton wheels because they were really the only enemy that reduced me to tooth-gnashing, controller-smashing rage.
 
Those fleshy dog things in the Gutter. Their attack animation and hitbox is way out of whack.
 
Lesser Sanctuary Guardians. It's like fighting two Artoriases at the same time, or two Manuses, or two Kalameets. I couldn't do it. It's the only fight I just ran away from. I couldn't defeat them in a fair fight. 2crazy4me.

Those fleshy monsters are out of whack. At least at that point in the game, one would have a bow and some poisoned arrows along with a shield in 90-100% physical damage defense.

Although I did mention a couple bosses, I would like to bring up one normal enemy, the mages in the Shrine of Amana. Fans don't call it Touhou Swamp for nothing, especially if you have to deal with other enemies such as Archdrake Clerics (who hit hard with their maces) and Shrine Aberrations (those lizard-like hollows). The next area, the Crypt, has it in the form of those ghostly swordsman. One isn't too bad but when you reach a certain area, you better find the exit since some hollows love to spawn and hit a bell, which summons a few of said swordsmen.

I actually loved the ghost swordsmen in the crypt. They were threatening and dangerous without feeling too cheap. Was a nice moveset change from the normal, boring DS2 "overhead swing overhead swing tracking sweep" humanoids too.
 
I've always dreaded taking on the skeleton wheels of sadness and despair. They're the one thing that standing between me and Pinwheel, so dying to them is pretty disheartening.

Manus from the DLC is probably the most difficult boss in the game for me. He hits both fast and hard. Plus I find myself mixing up the estus flask and silver pendant during that fight. Quite frustrating when that happens.
 
Skeleton Wheels so far. I haven't played 2 yet, but so far, Skeleton Wheels have been the biggest annoyance/cheapest enemy. Blowdart Snipers didn't give me too many problems, but I had gotten my hands on a Spider Shield by then.
