The joke was Jack wanted to know what his 3000th (yes, this was his 3000th tweet in 2012, his account was four years old by this point) should be. Ray, as a joke, tweeted to him suggesting something MLP related, laughed, and went on as usual.

Jack blocked him, and reported Ray's Twitter account as a spam bot. Mike's intern arc was even more funny because Jack demanded a "no pony policy" in light of Mike becoming intern for the AH crew with Ray and Michael rejoicing as the three of them would likely discuss MLP to Jack's frustration.
