Some roles are derived from actual
sex differences (eg, there is no society where men give birth and breastfeed children) but traditionally the word 'gender' has been used to describe behaviors and attitudes related to men and women that are
not tied to physical sex differences. So:
Sex = boys have penises
Gender = boys wear blue
The lines can get blurry or controversial in some areas: the historical trend has unfortunately been to assume that gender differences must be based on physical sex differences (eg, women can't go into higher education, because it will upset their wombs and they will sicken and die), and then there has been backlash to that which goes too far in the opposite direction (eg, insisting that difference in upper body strength between men and women is due to sexism). But that's the basic idea.
To make things more confusing, TRAs first hijacked the idea of 'gender is a social construct' (true!) and then when that didn't satisfy the autogynephiles enough, they started insisting that
sex differences are a social construct as well (not true!). Hence all the ridiculous sperging about 'muh intersex' and 'doctors just randomly assign sex at birth, no one really knows!'
lmaoooo he thinks women are mean to him because women are jealous and bitchy, when actually they don't like him because they can tell he is a creepy troon

I know these guys are delusional and hate women, but somehow they always manage to surprise me.
